亚麻胚性细胞悬浮培养体系的建立和影响因素

摘 要:

亚麻品种‘双亚五号’的胚性愈伤组织诱导最佳培养基为MB (MS无机盐加B5维生素)+1.0 mg·L-1 2,4-D; 细胞初始悬浮培养的最佳培养基为MB+0.2 mg·L-1 6-BA; 细胞继代悬浮培养的最佳培养基为改良的MB (NH4NO3 减半, KNO3 加倍)+0.02 mg·L-1 2,4-D+0.2 mg·L-1 6-BA; 适宜的蔗糖量为50 g·L-1; 适宜的继代接种量为1.0~1.5 g·(25 mL)-1; 继代间隔为5~7 d。

Establishment and Influence Factors of Embryogenic Cell Suspension Lines of Linum usitatissimum L. in vitro

Abstract:

The optimum induction medium for the embryogenic callus of Linum usitatissimum ‘Shuangya No. 5’ was MB (MS inorganic salts supplemented with B5 vitamins)+1.0 mg·L-1 2,4-D. The optimum medium for initial culture of suspension cells was liquid MB supplemented with 0.2 mg·L-1 6-BA. The suspension cells were cultured on modified MB liquid medium NH4NO3 halved, KNO3 doubled) supplemented with 0.02 mg·L-1 2,4-D+0.2 mg·L-1 6-BA for subculturing. The suitable sucrose concentration was 50 g·L-1. The optimal inoculative densities were 1.0–1.5 g·(25 mL)-1, and the subculture period was 5–7 d.
